Abstract   The evolution of cancer through a series of progressive steps discussed in the rat liver model baaed on the multi-hit multi-step hypothesis has initiation, promotion and progression stepa Base on the rat hepatocarcinogenesis model, cancer can be prevented by e#liminating exposure to carcinogens and inhibiting the promotion step. The anti#-initiation potential of diallyl sulfide (DAS), major component of garlic, in hepatocarcinogeneeis of rat was studied by many investigators. However, the anti- promotive potential of DAS is not yet well known. P-glycoprotein (PGP) is controlled and produced by MDR (multidrug resistance) gene. PGP functions as an ATP dependant, non-specific toxic substance efflux pump. In normal tiesue it may increase cellular resistance. It is possible that PGP acts as a scavenging agent, as in the acavenging mechanism of Wattenberg, and DAS has anti-promotion activity through PGP. The anti-promotive potential of DAS on the development of preneoplastic hepatic enzyme altered foci related with PGP was examined in an in vivo mid-term assay system of Ito. The antipromotive potential of DAS was scored by comparing the number of glutathione S-tranaferase placental form positive foci (GST-P foci) and area (96) of the foci per tissue aection of liver uaing image analyzer. Using anti-P-glycoprotein monoclonal antibody (JSB-1 clane) immunoreactivity of DEN and/or DAS treated rat liver were examined and image analysis of the immunoreactivity was done for quantitative results as area (R). The results of this study suggest that DAS has anti-yromtive potential against DEN induced hepatocarcinogeneeis in rat and DAS has anti-promotion activity through PGP. Further studies of antipromotion effect of PGP productive substance and inhibition of anti-promotive activity of DAS by anti-PGP substance might be follow.
